Summary This present study was conducted to shed a light on the efficacy of natural irrigants (Chitosan and Miswak) versus traditional irrigant. Seventy-five human freshly extracted single-rooted anteriors and premolars were selected free from any cracks or fracture line and inspected under light microscope. All the teeth were inserted inside sterilization packs and autoclaved for 20 minutes at 121°C and 15 PSI pressure then bacterial suspension was prepared and inoculation of root canals with Enterococcus faecalis The bacteria were incubated for 3 weeks in order to insure a matured biofilm that fully covered the canal walls as well as the lumen of dentinal tubules. All teeth were instrumented and subjected to different irrigating solutions. Group I was irrigated with 12.5 percent Salvadora persica. Group II was irrigated hitocan Group III was irrigated with 5.25 percent Sodium hypochlorite. Group IV (positive control) was irrigated with Saline. After irrigation protocol, samples were taken using two sterile paper points. The colony forming units were counted (CFU),and then the roots were split longitudinally in the buccolingual plane to prepare the samples for SEM evaluation. Results of the current study showed that: Regarding antibacterial effect, the highest mean value was found in hitonon and Miswak, the lowest mean value
