Different methods for the determination of some antibacterial drugs, in bulk, dosage forms and in human plasma have been introduced in this thesis.
These drugs are ciprofloxacin hydrochloride, norfloxacin, metronidazole and tinidazole. & 99;& 104;& 114;omatographic part includes HPLC determination of some mixtures namely a binary mixture of ciprofloxacin and metronidazole in absence or in presence of acid degradation product of ciprofloxacin and a tertiary mixture of norfloxacin, metronidazole and tinidazole.
An UHPLC determination of the tertiary mixture and a stability indication determination of norfloxacin in presence of its acid induced degradation product was applied together with a simultaneous determination of ciprofloxacin in presence of its acid induced degradation product with monolithic column.
In addition to a determination of ciprofloxacin in presence of one of its metabolites after being synthesized, in spiked human plasma using UHPLC was proposed.
Also, the & 99;& 104;& 114;omatographic part includes UHPLC-MS/MS simultaneous determination of ciprofloxacin and metronidazole in spiked human plasma.
Spectroscopic part includes the simultaneous spectrophotometric determination of a binary mixture; namely ciprofloxacin-metronidazole in presence of ciprofloxacin acid degradation product using CLS, PCR and PLS chemometric methods and artificial neural networks. Also, the spectroscopic part includes spectrofluorimetric determination of
metronidazole and tinidazole separately applying quantum dots fluorescence quenching technique.
Statistical comparison between the results obtained by the proposed methods and reference methods was carried out.
Acid induced degradation products of ciprofloxacin and norfloxacin were prepared and their structure was verified. As well as metabolite synthesis, structure elucidation and docking study of ciprofloxacin and its metabolites on enzyme was carried out.
